Quality Control of total RNA

 

The Center for Medical Genomics strongly recommends that at least 4 to 6 biologically independent samples be used for each condition to be examined. We do not recommend pooling strategies. In some circumstances, pooling is useful to get enough RNA to use, but we still recommend at least 4 to 6 biologically independent pools.

 

Amount of total RNA needed for the GeneChip

       Standard protocol: 5-10 mg total RNA at > 0.75 mg/ml

       With consultation: 2 mg to 5 mg total RNA in < 8 ml volume (we can concentrate samples for additional charge)

       Special: requires Amplification. 0.05 mg to 1 mg total RNA. Additional charge. NOTE: Amplified samples work, but there is truncation of the 5Õ ends and increased variability; consider using a larger number of samples. CONSULT WITH CMG to discuss the experiment.

 

Quality Control measures

       Agarose gel

       Spectrum from 210 or 220 nm to 350 nm

 

Detailed sample and experimental information should be provided before the sample is brought to the CMG.
